A Study of Lectin Histochemistry on Normal Skin of Human, Rat, Rabbit and Guinea pig


Abstract
Lectins are plant-derived protein which bind specially to simple or complex carbohydrate group. As such, Lectin are useful tools in studying glycoprotein and glycolipid structure of cell surface.
The author investigated the binding of Lectin to normal skin of human, rat, rabbit and guinea pig. ¢¥¢¥Using the avidin biotin peroxidase complex(ABC) method, applicable to formalin fixed paraffin embedded tissue, the binding properties of eight Lectins were investigated; Concanavalin A (Con-A), Peanut agglutinin (PNA), Ricinus communis agglutinin I (RCA I), Soybean agglutinin (SBA), Dolichos biflorus agglutinin (DBA), Wheat germ agglutinin (WGA), Ulex europaeus agglutinin I (UEA I), Pinellia ternata agglutinin (PTA).
The result obtained are as follow:
1. On the horny cell layer, Con-A showed positive reaction to human and rats, Con-A and PNA showed positive reaction to guinea pig, Con-A, PTA and RCA I showed positive reaction to rabbits.
2. On the spinous cell layer and granular cell layer, Con-A, UEA I, PNA and RCA I showed positive reaction to human. Con .A, WGA and RCA I revealed positive reaction on the granular cell layer of guinea pig, WGA and RCA I revealed positive reaction on the granular cell layer of rats.
3. On the basal cell layer, PNA, DBA, Con-A and WGA showed positive reaction to human. The other experimental animals showed negative reaction.
Melanin containing kerationcyte bound PTA and WGA specifically in normal human epidermis.
4. On sebaceous gland, Con-A, RCA I, PNA and PTA showed positive reaction in human, Con-A, PTA, UEA I and RCA I showed positive reaction in rats, Con-A and PTA showed
;positive reaction in guinea rig, FNA, Con-A, WGA and RCA I showed positive reaction in rabbits.
5. Eccrine gland were existed in human and rats, but author are not found its in rabbits and guinea pig. On the glandular portion of human eccrine gland, SBA, DBA, WGA and RCA I showed positive reaction. Con-A, PTA, WGA and RCA I revealed positive reation on ductal portion of human eccrine gland. On the glandular portion of rat eccrine gland, WGA, RCA I, Con-A and PTA showed positive reaction, DBA, UEA I showed focal positive reaction. WGA and RCA I revealed positive reaction on ductal portion of rat eccrine gland.
6. Apocrine gland were only found in normal human skin. SBA, PNA, DBA and WGA bound the luminal border in apocrine gland. Con-A, PTA, RCA I and UEA I showed positive cytoplasmic reaction on apocrine gland.
7. PNA labelled the hair matrix on human skin. In rats, ConA, PTA and RCA I bound the hair shaft, PNA bound the hair matrix. In guinea pig, RCA I, Con-A and WGA bound the hair shaft, DBA bound the hair matrix. In rabbits, WGA bound the hair matrix.
The author also found that RCA I stained the endothelial cell in rabbits.
